Can i ground the amplifier to the usb or the console

In a portable, all grounds are connected together to form a contiguous path of return. This means you can ground perpherals like the screen, GC+, or audio amp basically anywhere that is convenient for your build. There isn't really a right or wrong location.
The same is true for voltage. You can tap it from anywhere along the voltage path to power whatever you need, so long as the wire/trace is thick enough. You don't need to run a wire directly from the regulators to each component, you can daisy chain matching voltage locations from board to board instead. Typically, people choose to run power and ground to the Wii and then use the Wii's many voltage and ground locations as a free breakout to power perpherals.

Stitches is right, you want a common ground for everything in your build.
Having your USB GND going to your console and the GND for your amp going to that as well is what you’d want to do.
Like stated above, all ground are connected together.
(Make sure they are all connected, don’t wire it like you mentioned in your question).

It's a battery with 2 ports a 12v dc port and a 5v USB port the amp runs off 5v but the console runs off 12v
Make sure both devices use the same Ground, but obviously supply different voltages to the console (12V) and the amp (5V). If you need to step down the voltage with a regulator (12 to 5V), then do that, what are you using for your battery management system (BMS)?
Yeah, just make sure that all of your GND is wired up touching each other in layman's terms.
